📌 Indications:

Ruxotor (Ruxolitinib) is prescribed for:

  • Myelofibrosis (MF): including primary MF, post–polycythemia vera MF, and post–essential thrombocythemia MF.

  • Polycythemia Vera (PV): in adult patients resistant or intolerant to hydroxyurea.

  • Graft-versus-Host Disease (GVHD): treatment of steroid-refractory acute and chronic GVHD.


💊 Pharmacology:

  • Ruxolitinib is a potent Janus-associated kinase (JAK1 & JAK2) inhibitor.

  • Overactive JAK-STAT signaling contributes to abnormal blood cell production, inflammation, and immune dysfunction.

  • By blocking this pathway, Ruxotor helps reduce splenomegaly, control blood counts, alleviate disease-related symptoms, and improve overall survival in patients with MF and PV.


💉 Dosage & Administration:

  • Myelofibrosis/Polycythemia Vera: Initial dose is individualized based on platelet count (generally 5–20 mg twice daily).

  • GVHD: 5 mg twice daily, with dose adjustment according to tolerance.

  • Tablets should be swallowed whole with water, with or without food.

  • Dose modifications are necessary in case of low blood counts or adverse effects.


⚠️ Precautions & Warnings:

  • Monitor for anemia, thrombocytopenia, and neutropenia.

  • Risk of serious infections (TB, herpes zoster, hepatitis B reactivation).

  • Long-term use may increase the risk of non-melanoma skin cancers.

  • Regular complete blood count (CBC) monitoring is essential.


❌ Contraindications:

  • Hypersensitivity to Ruxolitinib or formulation components.

  • Severe organ dysfunction without close monitoring.


🤰 Pregnancy & Lactation:

  • May cause fetal harm; avoid during pregnancy.

  • Breastfeeding is not recommended while on therapy.
